BCKDK: The Master Switch for Branched-Chain Amino Acid Breakdown
Branched-Chain Ketoacid Dehydrogenase Kinase (BCKDK) is a mitochondrial enzyme with a single, highly specific role in cells: phosphorylation and inactivation of the branched-chain alpha-ketoacid dehydrogenase (BCKDH) complex. BCKDH is the rate-limiting enzyme that drives the catabolic breakdown of the three branched-chain amino acids: leucine, isoleucine, and valine. BCKDK inactivates BCKDH and therefore serves as a master brake on BCAA catabolism. Mutations that result in a loss of BCKDH function cause maple syrup urine disease (MSUD). Hyperactivity of BCKDK resulting in elevated BCAA levels has been shown to be a risk factor in insulin resistance and heart failure. Therefore, inhibition of BCKDK is a promising strategy to drive BCAA catabolism and lower their abundance in metabolic disorders.

Background
The branched-chain alpha-ketoacid dehydrogenase complex (BCKD) is a key enzyme that regulates the valine, leucine and isoleucine catabolic pathways. This gene encodes a protein that is located in the mitochondrion and inactivates BCKD by phosphorylation. Several transcript variants encoding different isoforms have been identified for this gene.
