BIRC5: A Cancer-Specific Regulator of Life and Death
Baculoviral IAP Repeat Containing 5 (BIRC5), also known as Survivin, is the smallest member of the family of inhibitor of apoptosis proteins (IAPs). This molecule is critically important in cancer, where it performs two major functions. First, it can directly inhibit the activity of caspases to block apoptosis. Second, as part of the chromosomal passenger complex (along with AURKB), BIRC5 is important for regulating microtubule dynamics and thus proper cell division. Expression of BIRC5 is very high during embryonic development but silenced in most terminally differentiated adult tissues. Dramatic re-expression in almost all human cancers and near absence from normal adult tissues makes it a perfect, highly selective target for cancer therapy.

Background
This gene is a member of the inhibitor of apoptosis (IAP) gene family, which encode negative regulatory proteins that prevent apoptotic cell death. IAP family members usually contain multiple baculovirus IAP repeat (BIR) domains, but this gene encodes proteins with only a single BIR domain. The encoded proteins also lack a C-terminus RING finger domain. Gene expression is high during fetal development and in most tumors, yet low in adult tissues. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene.
