C9: The Pore-Forming Unit of the Complement System
Complement C9 is the final component of the complement cascade. Following insertion of the C5b-8 complex into the target membrane, C9 binds to its polypentapeptide binding site and undergoes a rapid polymerization event. 10-18 copies of C9 insert and polymerize into a large, ring-shaped pore, the Membrane Attack Complex (MAC). This pore disrupts the osmotic integrity of the cell, causing rapid lysis and death. In addition to its importance in killing bacteria, unregulated MAC formation on self-cells is a major contributor to tissue damage in diseases such as paroxysmal nocturnal hemoglobinuria (PNH) and atypical hemolytic uremic syndrome (aHUS).

Background
This gene encodes the final component of the complement system. It participates in the formation of the Membrane Attack Complex (MAC). The MAC assembles on bacterial membranes to form a pore, permitting disruption of bacterial membrane organization. Mutations in this gene cause component C9 deficiency.

Component of the membrane attack complex (MAC). MAC assembly is initiated by proteolytic cleavage of C5 into C5a and C5b. C5b binds sequentially C6, C7, C8 and multiple copies of the pore-forming subunit C9 (PubMed:22832194). About 20 C9 chains oligomerize to give rise to a huge beta-barrel that forms a 100 Angstrom diameter pore in target membranes (PubMed:26841934, 30111885).
