CA8: An Acatalytic Player in Cerebellar Function
Carbonic Anhydrase-Related Protein 8 (CA8) belongs to the alpha-carbonic anhydrase family of proteins but it lacks the histidine residues that coordinate a zinc ion, the active site of the carbonic anhydrase enzyme, which makes it catalytically inactive. It is postulated that it may have scaffolding protein or signaling modulator functions instead. This protein is highly expressed in the cerebellar Purkinje cells, which are specialized neurons responsible for motor control and learning. Mutations that inactivate CA8 function result in a type of congenital ataxia, suggesting that this acatalytic family member plays an essential non-catalytic role for normal Purkinje cell signaling and survival and is a critical target for neurobiological study.

Background
The protein encoded by this gene was originally called CA-related protein based on sequence similarity to other previously known carbonic anhydrase genes. However, this gene product does not have carbonic anhydrase activity (i.e. the catalysis of the reversible hydration of carbon dioxide). Nevertheless, the gene product still has a carbonic anhydrase designation based on obvious sequence identity with other members of the carbonic anhydrase gene family. Failure to detect transcription of the CA8 gene in the cerebellum of the lurcher mutant, a mouse that has a neurologic defect, points to a possible significant role for this acatalytic form of the protein. Mutations in this gene have been found to cause cerebellar ataxia, mental retardation, and dysequilibrium syndrome 3 (CMARQ3).
