CALM1 is the ubiquitous calcium sensor protein that regulates virtually all aspects of cell biology. The need for excellent tools to study calcium signaling in the myriad of cellular contexts is so fundamental and diverse it ranges from neurotransmission to cardiac arrhythmias and everything in between.

CALM1: The Ubiquitous Calcium Sensor

Calmodulin 1 (CALM1) is one of three genes (CALM1, CALM2, and CALM3) that encode the same, ubiquitously expressed, and truly indispensable calcium-binding protein, calmodulin. Calmodulin is a small (approximately 17kD) dumbbell-shaped protein that has four EF-hand motifs that bind calcium. When calcium binds to calmodulin, it induces a large conformational change in the protein that allows it to bind to (and regulate the activity of) hundreds of downstream effector proteins, including protein kinases (e.g., CaMKII), phosphatases (e.g., calcineurin), and ion channels. Calmodulin is the primary and most important transducer of calcium signals in all eukaryotic cells. Mutations in any of the calmodulin genes lead to severe, often life-threatening cardiac arrhythmias.

Background

This gene encodes one of three calmodulin proteins which are members of the EF-hand calcium-binding protein family. Calcium-induced activation of calmodulin regulates and modulates the function of cardiac ion channels. Two pseudogenes have been identified on chromosome 7 and X. Multiple transcript variants encoding different isoforms have been found for this gene.A missense mutation in the CALM1 gene has been associated with ventricular tachycardia.
