CHKA: A Key Enzyme in Membrane Synthesis and Cancer
Choline kinase alpha (CHKA) is a cytosolic enzyme that phosphorylates choline using ATP to produce phosphocholine. This is the first and committed step in the CDP-choline or Kennedy pathway, the predominant pathway for the de novo synthesis of phosphatidylcholine (PC), the most common phospholipid of eukaryotic cell membranes. Rapidly proliferating cancer cells have an enormous demand for new membranes and therefore often upregulate this pathway. CHKA is overexpressed in a broad spectrum of human cancers and its high activity is required not only to build membranes but also generates phosphocholine, which acts as a signaling molecule to promote cell growth.

Background
The major pathway for the biosynthesis of phosphatidylcholine occurs via the CDP-choline pathway. The protein encoded by this gene is the initial enzyme in the sequence and may play a regulatory role. The encoded protein also catalyzes the phosphorylation of ethanolamine. Two transcript variants encoding different isoforms have been found for this gene.

[Isoform 1]: Monomer; acetylation by KAT5 promotes dissociation of the homodimer and monomerization. (Microbial infection) Interacts with PI4KA/PI4KIIIalpha; CHKA bridges PI4KA/PI4KIIIalpha and hepatitis C virus (HCV) non-structural protein 5A (NS5A) and potentiates NS5A-stimulated PI4KA activity, which then facilitates the targeting of the ternary complex to the ER for viral replication.
