CKB: The Brain-Specific Creatine Kinase

Creatine Kinase, Brain (CKB), also known as CK-BB, is a cytosolic enzyme with high expression levels in brain tissue as well as smooth muscle and various cancers. It is one of three isoenzymes involved in the creatine-phosphocreatine “shuttle.” CKB specifically catalyzes the reversible transfer of a phosphate group from ATP to creatine, forming phosphocreatine, which serves as a rapidly mobilizable high-energy reserve. In the brain, the creatine-kinase/phosphocreatine system has been postulated to buffer ATP levels in neurons and astrocytes in areas with high synaptic activity. CKB is also known to be frequently overexpressed in cancer, where it is thought to contribute to the high energy demands of tumors.

Background

The protein encoded by this gene is a cytoplasmic enzyme involved in energy homeostasis. The encoded protein reversibly catalyzes the transfer of phosphate between ATP and various phosphogens such as creatine phosphate. It acts as a homodimer in brain as well as in other tissues, and as a heterodimer with a similar muscle isozyme in heart. The encoded protein is a member of the ATP:guanido phosphotransferase protein family. A pseudogene of this gene has been characterized.
