CLPP: The Proteolytic Engine of Mitochondrial Protein Quality Control
Caseinolytic peptidase P (CLPP) is the catalytic core of a family of proteases. In the mitochondrial matrix, it assembles with other subunits to form a barrel-shaped complex that non-covalently binds with the ATPase chaperone ClpX. The resulting ClpXP protease complex degrades misfolded, damaged, or redundant proteins in the mitochondria in a process that is critical for protein homeostasis (proteostasis). ClpXP is also the central initiator of the mitochondrial unfolded protein response (UPRmt) stress signaling pathway. Loss-of-function mutations in CLPP lead to Perrault syndrome, a rare genetic disorder characterized by hearing loss and ovarian dysfunction.

Background
The protein encoded by this gene belongs to the peptidase family S14 and hydrolyzes proteins into small peptides in the presence of ATP and magnesium. The protein is transported into mitochondrial matrix and is associated with the inner mitochondrial membrane.

Fourteen CLPP subunits assemble into 2 heptameric rings which stack back to back to give a disk-like structure with a central cavity. Component of the Clp complex formed by the assembly of two CLPP heptameric rings with two CLPX hexameric rings, giving rise to a symmetrical structure with two central CLPP rings flanked by a CLPX ring at either end of the complex.
