CSNK1E: A Master Regulator of the Circadian Clock

Casein Kinase 1 Epsilon (CSNK1E) is a serine/threonine protein kinase that is a part of the central negative feedback loop of the molecular circadian clock. It phosphorylates the PERIOD (PER) and CRYPTOCHROME (CRY) proteins, the core negative regulators of the clock, in the nucleus of the cell. This marks PER and CRY for degradation by the ubiquitin-proteasome system, which is one of the most important steps for timing the ~24-hour cycle of circadian gene expression. Mutations that enhance or reduce CSNK1E activity cause inherited human disorders of sleep timing, and altered CSNK1E expression has been implicated in depression and bipolar disorder.

Background

The protein encoded by this gene is a serine/threonine protein kinase and a member of the casein kinase I protein family, whose members have been implicated in the control of cytoplasmic and nuclear processes, including DNA replication and repair. The encoded protein is found in the cytoplasm as a monomer and can phosphorylate a variety of proteins, including itself. This protein has been shown to phosphorylate period, a circadian rhythm protein. Two transcript variants encoding the same protein have been found for this gene.
