CTPS1: The Key Enzyme in Pyrimidine Synthesis

CTP Synthase 1 (CTPS1) is an enzyme involved in nucleotide metabolism that facilitates the formation of cytosine triphosphate (CTP) from UTP in the pyrimidine synthesis pathway. In this process, CTPS1 utilizes glutamine as a nitrogen donor. CTP, one of the four nucleotides necessary for DNA and RNA synthesis, is also involved in phospholipid synthesis. CTPS1 is the primary isoform expressed in lymphocytes and rapidly dividing cancer cells and is thought to be crucial for T-cell proliferation and certain leukemias. CTPS1 activity is often found at filamentous cytoophidia, implicating it as having a non-metabolic role in regulating pyrimidine biosynthesis. This vital role in proliferation and pyrimidine synthesis makes CTPS1 an attractive target for anti-cancer and immunosuppressive drugs.
