CTSS: The Master Protease of Antigen Presentation
Cathepsin S (CTSS), a lysosomal cysteine protease, is highly expressed by professional antigen-presenting cells (APCs) including dendritic cells, B-cells and macrophages. Its primary role is to cleave the invariant chain (Ii) which is non-covalently bound to newly synthesized MHC class II molecules. Cathepsin S removes the last piece of Ii called CLIP, thereby making the peptide-binding groove accessible for antigenic peptides to be loaded onto the MHC-II molecule for presentation to CD4+ T-helper cells. The loading of peptide antigens onto MHC-II molecules is the rate-limiting step in the activation of T-helper cells, so inhibition of Cathepsin S is a logical approach to suppress autoimmune responses.
