CASP4: A Direct Sensor of Intracellular Bacteria

Caspase-4 (CASP4) and its close homolog Caspase-5 are human inflammatory caspases. Their murine ortholog is Caspase-11. Unlike the canonical NLRP3 inflammasomes that require expression of separate sensor proteins, Caspase-4 is able to directly recognize intracellular lipopolysaccharide (LPS) (the component of gram-negative bacterial outer membrane) when it is present in the cytosol of the host cell, and act as its own sensor protein. This direct recognition results in oligomerization and activation of caspase-4, which then cleaves Gasdermin D, resulting in pyroptosis, and also activates the NLRP3 inflammasome, promoting IL-1beta production. This critical anti-intracellular bacterial defense pathway is also a central driver of septic shock.

Background

This gene encodes a member of the cysteine proteases that plays important roles in apoptosis, cell migration and the inflammatory response. The encoded protein mediates production of pro-inflammatory cytokines by macrophages upon bacterial infection. Mice lacking the encoded protein are resistant to endotoxic shock induced by lipopolysaccharide. A 5-bp deletion encompassing a splice acceptor junction resulting in alternate splicing and a shorter non-functional isoform in certain mouse strains has been described.
