EPHX2: The Soluble Epoxide Hydrolase in Cardiovascular Regulation
The cytosolic enzyme Epoxide Hydrolase 2 (EPHX2), also known as soluble epoxide hydrolase (sEH), is a metabolizing enzyme that terminates the actions of epoxyeicosatrienoic acids (EETs). EETs are anti-inflammatory and vasodilatory lipid signaling molecules that are derived from arachidonic acid. By converting these cytoprotective EETs to their inactive diol forms, sEH has opposing effects to EETs. Inhibiting sEH increases tissue EET levels, leading to lower blood pressure, decreased inflammation, and diminished pain. In fact, sEH is a major therapeutic target for cardiovascular and inflammatory disorders.

Background
This gene encodes a member of the epoxide hydrolase family. The protein is predominantly located in the cytosol and peroxisomes. It has been shown to bind to specific epoxides and convert them to the corresponding dihydrodiols. Mutations in this gene have been associated with familial hypercholesterolemia. Alternatively spliced transcript variants have been described.
