GRK2: The Master Regulator of Beta-Adrenergic Receptor Desensitization
G protein-coupled receptor kinase 2, also known as beta-adrenergic receptor kinase 1 or βARK1, is a serine/threonine kinase which phosphorylates and desensitizes G protein-coupled receptors. Beta-adrenergic receptors in the heart are chronically stimulated during heart failure. This leads to an upregulation and over-activity of GRK2. GRK2 phosphorylates the activated beta-adrenergic receptors, which facilitates binding of β-arrestin and internalization of the receptor, effectively shutting down receptor signaling. This leads to a dampened heart response to adrenaline and contributes to a failure to maintain cardiac output. Inhibition of GRK2 is a highly desirable therapeutic target for the treatment of heart failure.

Background
This gene encodes a member of the G protein-coupled receptor kinase family of proteins. The encoded protein phosphorylates the beta-adrenergic receptor as well as a wide range of other substrates including non-GPCR cell surface receptors, and cytoskeletal, mitochondrial, and transcription factor proteins. Data from rodent models supports a role for this gene in embryonic development, heart function and metabolism. Elevated expression of this gene has been observed in human patients with heart failure and Alzheimer's disease.
