Background
GSDME-mediated pyroptosis is a distinct and increasingly recognized pathway of regulated cell death (RCD) that highlights a crucial intersection between the traditionally "silent" apoptotic process and the highly "inflammatory" pyroptotic cell death. While canonical and non-canonical pyroptosis primarily depend on inflammatory caspases (Caspase-1, -4, -5, -11) cleaving Gasdermin D (GSDMD), the discovery of Gasdermin E (GSDME, also known as DFNA5) as a substrate for apoptotic caspases has unveiled an alternative route to pyroptosis.
