This gene encodes a member of the interleukin-1 receptor-associated kinase protein family. Members of this family are essential components of the Toll/IL-R immune signal transduction pathways. This protein is primarily expressed in monocytes and macrophages and functions as a negative regulator of Toll-like receptor signaling. Mutations in this gene are associated with a susceptibility to asthma. Alternate splicing results in multiple transcript variants.
Protein class

Disease related genes, Enzymes, Metabolic proteins, Plasma proteins, Potential drug targets

Predicted location

Intracellular

Single cell type specificity

Cell type enhanced (Macrophages, Kupffer cells, Microglial cells, monocytes, Adipocytes)

Immune cell specificity

Group enriched (non-classical monocyte, intermediate monocyte, classical monocyte, neutrophil, basophil, myeloid DC)

Cell line specificity

Cell line enhanced (ASC diff, ASC TERT1, HEL, HL-60, NB-4, THP-1, U-937)

Interaction

Monomer (PubMed:33238146). Homodimer; disulfide-linked (PubMed:33238146). May interact with IRAK4 (when phosphorylated) (PubMed:33238146). Interacts (when phosphorylated at Ser-110) with PIN1 (via WW domain) in response to IL33-mediated (but not TLR4 ligand LPS) dendritic cell stimulation (PubMed:29686383).
