NPR2: The Critical Receptor for Bone Growth

A single-pass transmembrane receptor also known as guanylate cyclase B, NPR2 is highly expressed in the growth plates of long bones. Activation of NPR2 by its endogenous ligand, C-type natriuretic peptide (CNP) leads to production of the second messenger cGMP. The CNP-NPR2-cGMP pathway is the most robust endogenous stimulator of endochondral ossification, the cellular process responsible for the growth of bones in length. Loss-of-function mutations in the NPR2 gene result in Acromesomelic Dysplasia, Maroteaux type (AMDM), a form of dwarfism, further characterizing NPR2 as a hot-spot for skeletal diseases.

Background

This gene encodes natriuretic peptide receptor B, one of two integral membrane receptors for natriuretic peptides. Both NPR1 and NPR2 contain five functional domains: an extracellular ligand-binding domain, a single membrane-spanning region, and intracellularly a protein kinase homology domain, a helical hinge region involved in oligomerization, and a carboxyl-terminal guanylyl cyclase catalytic domain. The protein is the primary receptor for C-type natriuretic peptide (CNP), which upon ligand binding exhibits greatly increased guanylyl cyclase activity. Mutations in this gene are the cause of acromesomelic dysplasia Maroteaux type.
