PCYT1A: The Rate-Limiting Enzyme for PC Synthesis

CTP: Phosphocholine Cytidylyltransferase Alpha (PCYT1A) is the rate-limiting enzyme in the de novo pathway of phosphatidylcholine (PC) synthesis. PC is the most abundant phospholipid in cell membranes. PCYT1A transfers the cytidylyl group from CTP to phosphocholine to generate CDP-choline. This is the rate-limiting step of the entire pathway. It is under tight control and is only active when it senses that more membrane is needed. It does so by binding to PC-deficient membranes like the nuclear envelope and the endoplasmic reticulum (ER). The enzyme exists as an active membrane-bound form and an inactive soluble dimer that resides in the nucleus. It is only active as a membrane-bound heterodimer. In this way, the enzyme is a key metabolic sensor for membrane biogenesis.

Background

This gene belongs to the cytidylyltransferase family and is involved in the regulation of phosphatidylcholine biosynthesis. Mutations in this gene are associated with spondylometaphyseal dysplasia with cone-rod dystrophy. Alternatively spliced transcript variants have been found for this gene.
