PDE10A: A Key Regulator of Striatal Signaling

Phosphodiesterase 10A (PDE10A) is a dual-substrate phosphodiesterase that can hydrolyze both cyclic AMP (cAMP) and cyclic GMP (cGMP), the two most important second messengers in the cell. PDE10A expression in the body is very limited, with the highest levels being expressed in the medium spiny neurons of the striatum in the brain. This region of the brain is a key site for the integration of motor control, motivation and cognition. By regulating the intracellular concentration of cAMP/cGMP, PDE10A is a key modulator of the direct and indirect pathways of signaling in the striatum. Abnormal signaling in these pathways is a fundamental feature of neuropsychiatric diseases such as schizophrenia and Huntington's disease, making PDE10A an attractive target for therapeutic intervention.

Background

The encoded protein is a member of the family of cyclic nucleotide phosphodiesterases. These enzymes are important signal transducers which regulate the intracellular concentration of the second messengers, cyclic nucleotides. The encoded enzyme is able to hydrolyze both cAMP and cGMP to the corresponding nucleoside 5' monophosphate, but binds cAMP with higher affinity and is a more efficient phosphodiesterase with cAMP as substrate. Alternatively spliced transcript variants have been described for this gene.
