PDE5A: The cGMP-Specific Target of Sildenafil

Phosphodiesterase 5A (PDE5A) is an enzyme that selectively degrades the second messenger cyclic GMP (cGMP) in a wide variety of tissues. It is perhaps best known for its expression in the corpus cavernosum of the penis, as well as in the smooth muscle cells of the pulmonary vasculature, where it plays an essential role in modulating the physiological effects of nitric oxide (NO). In these tissues, NO signaling triggers an increase in cGMP that induces smooth muscle relaxation and vasodilation. PDE5A functions as the "off switch" by degrading cGMP to turn off this signal. Inhibitors of PDE5A, like sildenafil (Viagra), block this cGMP breakdown, leading to persistently high cGMP levels that cause exaggerated vasodilation, which has therapeutic utility for erectile dysfunction and pulmonary arterial hypertension.

Background

Glutamic-oxaloacetic transaminase is a pyridoxal phosphate-dependent enzyme which exists in cytoplasmic and inner-membrane mitochondrial forms, GOT1 and PDE5A, respectively. GOT plays a role in amino acid metabolism and the urea and tricarboxylic acid cycles. The two enzymes are homodimeric and show close homology. Two transcript variants encoding different isoforms have been found for this gene.
