Background

The 26S proteasome is a multicatalytic proteinase complex with a highly ordered structure composed of 2 complexes, a 20S core and a 19S regulator. The 20S core is composed of 4 rings of 28 non-identical subunits; 2 rings are composed of 7 alpha subunits and 2 rings are composed of 7 beta subunits. The 19S regulator is composed of a base, which contains 6 ATPase subunits and 2 non-ATPase subunits, and a lid, which contains up to 10 non-ATPase subunits. Proteasomes are distributed throughout eukaryotic cells at a high concentration and cleave peptides in an ATP/ubiquitin-dependent process in a non-lysosomal pathway. An essential function of a modified proteasome, the immunoproteasome, is the processing of class I MHC peptides. This gene encodes one of the ATPase subunits, a member of the triple-A family of ATPases that have chaperone-like activity. This subunit may compete with PSMC2 for binding to the HIV tat protein to regulate the interaction between the viral protein and the transcription complex. A pseudogene has been identified on chromosome 9.

We used the Mouse Anti-PSMC3 Recombinant Antibody for flow cytometry on Jurkat cells. The antibody produced a clear signal with low background, and the staining pattern was consistent across replicates. It required minimal adjustment to our standard FC protocol. Overall, it performed well and proved to be a reliable reagent for PSMC3 detection.

Mouse Anti-NHP PSMC3 Recombinant Antibody (clone OTI1C10) (CAT#: VS-1024-XY420)
In our Western blot tests, we used the Mouse Anti-NHP PSMC3 Recombinant Antibody with lysates from COS-7 cells. The antibody showed a strong, specific band at the expected size and minimal non-specific binding. It integrated smoothly into our workflow and produced consistent results. A dependable option for WB applications in non-human primate samples.

Mouse Anti-PSMC3 Recombinant Antibody (clone 1G10B7) (CAT#: VS7-0425-WR705)
We applied the Mouse Anti-PSMC3 Recombinant Antibody in Western blot using lysates from HeLa cells. The antibody produced a distinct band at the correct molecular weight with low background. It worked well under standard conditions and didn’t require much optimization. Overall, it's a reliable reagent for WB analysis.
