PTS: A Key Metabolic Regulator in Genetic Disorders Cancer and Microbial Pathogenesis

PTS serves as a critical enzyme in the tyrosine biosynthesis pathway, with significant implications across multiple pathological conditions. In metabolic disorders, its dysregulation contributes to hereditary tyrosinemia type I through toxic metabolite accumulation, causing severe liver dysfunction and neurological complications. The enzyme's role in phenylalanine metabolism links it to phenylketonuria (PKU), where impaired activity leads to neurocognitive deficits. Oncological research reveals its overexpression in certain malignancies, promoting tumor proliferation through enhanced aromatic amino acid production supporting protein synthesis. In neurodegenerative diseases, abnormal activity correlates with dopamine metabolism disturbances observed in Parkinson's pathology. Microbial pathogenesis studies highlight its essentiality in bacterial folate synthesis, making it a potential target for antimicrobial development against drug-resistant strains. Recent investigations also demonstrate involvement in inflammatory bowel diseases through gut microbiome modulation of aromatic amino acid availability. The multifaceted roles in fundamental metabolic pathways position this target as particularly valuable for therapeutic intervention in conditions where amino acid imbalance, proliferative signaling, or microbial pathogenesis drive disease progression. Small molecule inhibitors and genetic modulation approaches show particular promise in preclinical models addressing these diverse mechanisms.

Alternative Names

Complement PTS; 6-Pyruvoyltetrahydropterin Synthase; PTP Synthase; PTPS; 6-Pyruvoyl Tetrahydrobiopterin Synthase; EC 4.2.3.12

Background

The enzyme encoded by this gene catalyzes the elimination of inorganic triphosphate from dihydroneopterin triphosphate, which is the second and irreversible step in the biosynthesis of tetrahydrobiopterin from GTP. Tetrahydrobiopterin, also known as BH(4), is an essential cofactor and regulator of various enzyme activities, including enzymes involved in serotonin biosynthesis and NO synthase activity. Mutations in this gene result in hyperphenylalaninemia.

Mouse Anti-PTS Recombinant Antibody (clone 39G11) (CAT#: MOB-1933z)
The Mouse Anti-PTS Antibody worked reliably in ELISA with recombinant protein samples, showing good sensitivity at 1:2000 dilution. We also tested it in WB using HEK293 lysates, where it detected a clean band at the expected size. While it performed consistently across batches, some background appeared in tissue homogenates unless blocking time was extended. A solid choice for basic detection assays.

Recombinant Mouse anti-Human PTS Monoclonal antibody (BU3C3) (CAT#: MOB-1725CT)
The Recombinant Mouse anti-Human PTS antibody performed well in Western blotting with HeLa and HepG2 cell lysates, showing a clean ~50kDa band at 1:1000 dilution. While the signal was strong, we noticed slight non-specific binding in mouse liver homogenates that required extended blocking. It worked consistently across multiple lots, though optimal dilution varied slightly between cell types. The antibody hasn't been tested in IP applications yet, but performs reliably for basic WB needs. Some optimization may be needed for tissue samples.

Rabbit Anti-PTS (pS19) Recombinant Antibody (clone R05-7G8) (CAT#: VS3-WK2041)
The Rabbit Anti-PTS antibody worked well in WB with stimulated HEK293T lysates, specifically detecting the phosphorylated form at ~50kDa. Minimal background was observed in brain tissue homogenates after optimizing blocking conditions. While it showed good specificity for the phosphorylated epitope, the signal intensity varied slightly between stimulation timepoints. Reliable for basic phospho-protein detection when proper controls are included.
