This product is a recombinant mouse anti-Dengue virus-2 monoclonal antibody. This antibody specifically binds to the domain III of dengue virus envelope glycoprotein. This monoclonal antibody is capable of specifically binding and neutralizing dengue virus. This antibody product can be potentially used in the research of Dengue virus-2 infection.
Figure 1 Virus neutralization and mode of action of 9F12.
Pre- and post-adsorption assay. Pre-incubated mixtures of DENV-2 with mAb 9F12 (open squares) or 4G2 (open circles) were dispensed onto Vero cell monolayers at 4 °C for 1 h in the pre-adsorption assay. The virus antibody mixture was replaced with diluted virus in the post-adsorption assay, followed by washing and addition of either mAb 9F12 (filled squares) or 4G2 (filled circles).

The assay was carried out in triplicate and the values displayed represent the average. Error bars, sd.

Figure 3 Epitope mapping.
Flow cytometry histograms of mAbs binding to yeast expressing wild-type or mutant domain III. The following antibodies were used as negative and positive staining controls, respectively, for each of the indicated yeast based on data from a prior publication: wild-type, WNV E16 and 3H5-1; K305E, 1A1D-2 and 3H5-1; K307C, 1A1D-2 and 3H5-1; K310E, 1A1D-2 and 3H5-1; P384A, 3H5-1 and 5A2-7; G330D, 6B6-10 and 3H5-1. In each case, staining with 9F12 is shown in red. The data are representative of three independent experiments.

Figure 4 ELISA results of the antibody.
Elisa for C9 IgG (30 nmol/L) against DENV2 whole virus particles competed with increasing concentrations of mouse 9F12 and 3H5 IgG. C9 IgG binding was detected with an anti-human horseradish peroxidase (HRP) conjugate.
