Recombinant Mouse Antibody (15C9) is capable of binding to Dabigatran, expressed in Chinese Hamster Ovary cells (CHO). The dabigatran binding site of the antibody is located in a concave region at the interface of the variable domains.
Figure 1 aDabi-Fab binds to dabigatran and reverses its effect in vitro.
(B) Immobilized mouse antibodies were analyzed for binding to a hapten-peroxidase conjugate, which was competed off by increasing concentrations of soluble dabigatran. As a representative example, 2 dabigatran-specific antibodies together with a negative control, mAb, are shown. Complete inhibition is observed at a dabigatran time interval. Each PRP sample was incubated with 5mM CaCl2 and 3 mg/mL aDabi-Fab (final concentration). SFLLRN (1-5mM final concentration, LOXO [1mM], Dossenheim, Germany) was used as a positive control.
