Recombinant Human monoclonal antibody expressed in CHO binding to Human HSP90. The anti-HSP90 antibody is capable of recognizing cell surface HSP90 with high sensitivity, and hence is capable of detecting cell surface HSP90 of living cell in FACS analysis. Because of its capability to specifically recognize cell surface HSP90 with high sensitivity, the antibody can be used, for example, for preparing the vaccine, for delivering a substance possessing a biological activity, for an adjuvant that enhances a function of a vaccine, a tumor antigen antibody or an anti-tumor cell antibody, and in a method of specifically detecting a cell expressing cell surface HSP90.

Western blot analysis was performed by loading 0.1 µg (Lane 1), 0.3 µg(Lane 2), and 0.6 µg (Lane 3) recombinant human HSP90 onto a 12% Tris-HCl polyacrylamide gel. Proteins were transferred to a CN membrane and blocked with 5% skim milk for at least 1 hour. Membranes were probed with Anti-Human HSP90 Therapeutic Antibody (TAB-368CL) at a dilution of 1:500 overnight at 4°C on a rocking platform. Then probed with Goat anti-Mouse IgG HRP secondary antibody at a dilution of 1:2,000 for one hour. Chemiluminescent was detected.

Dot Blot analysis of TAB-368CL was performed by coating with human HSP90 antigen.
TAB-368CL incubation concentration: 2 ng/μL.

ELISA analysis of TAB-368CL was performed by coating with recombinant human HSP90 antigen. Then blocked with BSA, and incubated with Anti-Human HSP90 Therapeutic Antibody (TAB-368CL). The HRP-conjugated Goat anti-Mouse IgG as a secondary antibody. Detection was performed using TMB substrate and stopped with sulfuric acid. The absorbances were read on a spectrophotometer at 450 nm.
