Recombinant Anti-EPCAM × Anti-CD3 Bispecific Antibody (Tandem Diabody) is a bispecific fusion protein with four binding sites, two of which bind to antigens of one kind of cells and the other two to the other kind of cells. Tandem Diabodies do not carry Fc domains, are smaller than whole IgGs or IgG-derived BsAbs but larger than tandem scFvs. Therefore, they have a shorter serum half-life than IgGs but remain longer in the circulation compared with tandem scFvs. This BsAb is capable of targeting the tumor and recruiting T effector cells via binding to the CD3 subunit of the T cell receptor complex. It was designed for the treatment of EPCAM positive epithelial cancer.

Potent Lysis Of Epithelial Cells
In our co-culture system with epithelial tumor cells and PBMCs, this Tandem Diabody induced very potent cell lysis. Its ability to recruit T cells to EpCAM positive targets is remarkable, even at relatively low effector-to-target ratios in our experiments.

Stable Tetravalent Format
We were concerned about the stability of antibody fragments, but this Tandem Diabody format is quite robust. It maintained its binding activity after freeze-thaw cycles, and the tetravalent binding provided strong signals in our FACS analysis of tumor cells.
