The antibody can be specifically bound with the A-beta oligomer, effectively inhibit the fibrosis aggregation of A-beta and obviously alleviate the toxic effect of the A-beta on cells.
Figure 1 In vitro characterization of anti-O antigen mAbs.
ScFv antibodies were transferred to nitrocellulose membrane and probed with 9E10 antibody. Bound antibodies were detected using a HRP-conjugated goat anti-mouse and visualized with DAB.
Wang, X. P., Zhang, J. H., Wang, Y. J., Feng, Y., Zhang, X., Sun, X. X., ... & Zhao, M. (2009). Conformation-dependent single-chain variable fragment antibodies specifically recognize beta-amyloid oligomers. FEBS letters, 583(3), 579-584.
Figure 2 In vitro characterization of anti-O antigen mAbs.
Aggregation of A42 was monitored by dot-blot. Aliquots of samples were applied to nitrocellulose membrane (0.12 μg/dot) at the time indicated and then probed with 4G8 and scFv antibodies, respectively.

Figure 3 Effects of scFv antibodies on fibrillation of Aβ42.
The kinetics of 10 lM Aβ42 fibril formation was monitored by ThT fluorescence in the absence or presence of W20 at different concentrations, respectively. The experiment was performed as indicated in Materials and methods section.

Figure 4 Effects of scFv antibodies on Aβ42-induced cytotoxicity.
Aliquots of Aβ42 preincubated with or without scFv's at indicated times were added to wells coated with SH-SY5Y cells. The final concentration of both Aβ42 and scFv was μlM. After an additional two-day incubation of cells and samples, the viability of the cells was measured using both MTT toxicity assay
