Anti-BCL11B Antibody Prodrug, Protease Activated (25B6)

The Anti-BCL11B Antibody Prodrug, Protease Activated (25B6) is an antibody prodrug comprised of a recombinant rabbit antibody targeting human BCL11B, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to BCL11B protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to BCL11B effectively. BCL11B (B-cell CLL/Lymphoma 11B) is a zinc finger transcription factor that acts as a tumor suppressor in T-cell malignancies. Loss-of-function mutations are frequently found in T-cell Acute Lymphoblastic Leukemia (T-ALL), where its inactivation removes a key transcriptional brake on T-cell proliferation and differentiation.

- This gene encodes a C2H2-type zinc finger protein and is closely related to BCL11A, a gene whose translocation may be associated with B-cell malignancies. Although the specific function of this gene has not been determined, the encoded protein is known to be a transcriptional repressor, and is regulated by the NURD nucleosome remodeling and histone deacetylase complex. Four alternatively spliced transcript variants encoding distinct isoforms have been found for this gene.
