The anti-CEACAM8 pH-dependent antibody (VS-0825-YC78) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CEACAM8. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CEACAM8 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CEACAM8 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CEACAM8 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CEACAM8 is a GPI-anchored cell surface protein expressed primarily on granulocytes. It is involved in cell adhesion and is used as a marker for certain types of myeloid cells.
