The anti-CX3CL1 pH-dependent antibody (VS-0825-YC88) targets human CX3CL1 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-CX3CL1 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced CX3CL1 clearance. The antibody operates in a cyclical manner, binding CX3CL1 in the extracellular fluid and releasing it in acidified endosomes. Human CX3CL1, also known as Fractalkine, is a chemokine that exists in both soluble and membrane-bound forms.
