The anti-IFNA1 pH-dependent antibody (VS-0825-YC163) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human IFN-alpha. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for IFN-alpha under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from IFN-alpha at pH 6.0. This pH-sensitive antibody retains its binding affinity for IFN-alpha on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human IFN-alpha, or Interferon-alpha, is a cytokine that is a key component of the innate immune response to viral infections. It is a potent antiviral agent and is used therapeutically for certain viral infections and cancers.
