The anti-IL17F pH-dependent antibody (VS-0825-YC177) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to both human and cynomolgus IL17F. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for IL17F under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from IL17F at pH 6.0. This pH-sensitive antibody retains its binding affinity for IL17F on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human and cynomolgus monkey IL17F are cytokines of the IL-17 family that play a role in inflammation and host defense. They share a high degree of homology and serve similar functions, making the cynomolgus monkey a valuable model for studying IL-17-related diseases.
