The anti-IL33 pH-dependent antibody (VS-0825-YC197) targets human Interleukin 33 (IL-33) in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-IL33 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced IL33 clearance. The antibody operates in a cyclical manner, binding IL-33 in the extracellular fluid and releasing it in acidified endosomes. Human Interleukin-33, or IL-33, is a cytokine that is a key player in the Th2 immune response. It is a potent inducer of allergic inflammation and is implicated in the pathogenesis of asthma.
