The anti-Integrin α4β7 pH-dependent antibody (VS-0825-YC225) targets monkey Integrin α4β7 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-Integrin α4β7 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced Integrin α4β7 clearance. The antibody operates in a cyclical manner, binding Integrin α4β7 in the extracellular fluid and releasing it in acidified endosomes. Integrin α4β7 is a heterodimeric cell surface receptor that is crucial for the homing of lymphocytes to the gut-associated lymphoid tissue. It is a major therapeutic target for inflammatory bowel diseases, such as Crohn's disease and ulcerative colitis.
