The anti-Integrin αVβ3 pH-dependent antibody (VS-0825-YC365) targets Integrin αVβ3 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-Integrin αVβ3 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced Integrin αVβ3 clearance. The antibody operates in a cyclical manner, binding Integrin αVβ3 in the extracellular fluid and releasing it in acidified endosomes. Integrin αVβ3 is a cell surface receptor for extracellular matrix proteins like vitronectin and fibronectin. It is a key mediator of cell adhesion and migration, and its expression is critical for angiogenesis and cancer progression.
