The anti-TGFB pH-dependent antibody (VS-0825-YC325) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to both human and mouse TGFB. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for TGFB under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from TGFB at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. TGFB (Transforming Growth Factor Beta) is a multifunctional cytokine that acts as a master regulator of cell proliferation, differentiation, and apoptosis. It has complex and context-dependent roles, functioning as both a tumor suppressor and a pro-tumorigenic factor.
