The anti-VEGFC pH-dependent antibody (VS-0825-YC354) targets human VEGFC in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-VEGFC acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced VEGFC clearance. The antibody operates in a cyclical manner, binding VEGFC in the extracellular fluid and releasing it in acidified endosomes. Human VEGFC (Vascular Endothelial Growth Factor C) is a growth factor that is a key regulator of lymphangiogenesis. It plays a critical role in the development of lymphatic vessels and is implicated in the lymphatic metastasis of cancers.
