The anti-Wnt pH-dependent antibody (VS-0825-YC361) targets human Wnt in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-Wnt acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced Wnt clearance. The antibody operates in a cyclical manner, binding Wnt in the extracellular fluid and releasing it in acidified endosomes. Wnt proteins are a family of secreted signaling proteins that regulate the Wnt signaling pathway. This pathway is crucial for embryonic development and tissue homeostasis, and its dysregulation is a major driver of many cancers.
