The APC-iFluor 700 antibody labeling kit is a convenient and efficient option for labelling antibodies or proteins with the red Laser-excitable XFD750 dye. It is important to note that impure antibodies or those stabilized with BSA or gelatin may not label well with this kit, and purification kits are available from creative biolabs to address this issue.

  • Procedure Steps
  • Performing an antibody-MTA reaction with approximately 100 µg of each component:
    1. Combine the antibody working solution with the MTA in a vial and mix thoroughly by pipetting or vortexing.
    2. Allow the antibody-MTA mixture to incubate at room temperature for 30-60 minutes. It can be rotated or shaken for a longer period if desired.
    Conjugating the antibody with APC-iFluor 700 with approximately 100 µg of each component:
    1. Prepare FOL-Activated APC-iFluor 700 solution by adding 50 µL of ddH2O to the vial and mix well.
    2. Combine the entire FOL-Activated APC-iFluor 700 solution with the antibody-MTA mixture and rotate the mixture for 1 hour at room temperature.
    3. The antibody-APC-iFluor 700 conjugate is now ready to use. If not using immediately, the conjugate may need to be diluted with a buffer of your choice.

  • This product is intended for research purposes only and is not intended for use in human, therapeutic, or diagnostic applications.
  • Kit Storage
  • Upon arrival, the kit should be stored at 4°C for optimal stability for up to six months. MTA can also be stored at -20°C if preferred. It is important not to freeze FOL-Activated APC-iFluo 700 or raction buffer. Prior to use, ensure all components are properly warmed and briefly centrifuged. Prepare the necessary solutions promptly before beginning the conjugation process. The following SOP provides an example for labeling goat anti-mouse IgG antibody.
  • Conjugate Storage
  • The antibody conjugate should be kept at a concentration of> 0.5 mg/mL with a carrier protein present (e.g., 0.1% bovine serum albumin). The antibody-APC-iFluor 700 conjugate solution can be stored at 4 °C for up to two months and protected from light to prevent significant changes. For extended storage, the antibody-APC-iFluor 700 conjugates can be lyophilized and stored at ≤ -20°C.
