  • Procedure Steps
  • Assay Procedure
    Equilibrate all materials and prepared reagents to room temperature before use.
    1. Add 1 µL of modifier per 10 µL of antibody to be labeled and mix gently.
    2. Remove the cap from the vial of fluorescent conjugation mixture and drop the antibody sample (with modifier) ​​directly onto the lyophilized material. Resuspend by gently aspirating and redispensing the liquid once or twice using a pipette.
    3. Cap the vial and incubate at room temperature (20-25°C) in the dark for 3 hours. Longer incubation times (e.g., overnight) have no negative effect on the conjugation.
    4. After 3 hours (or longer) of incubation, add 1 µL of quencher per 10 µL of antibody and mix gently. The conjugate is ready for use after 30 minutes. The conjugate does not require purification.

  • Recommended pre-conjugation buffer components and conditions:
    The antibody should be a purified antibody.
    Antibody concentration can be 0.5-1 mg/mL.
    pH can be 6.5-8.5.
    Amine free buffer (e.g. MES, MOPS, HEPES, PBS)
    Non-buffering salts (e.g. sodium chloride)
    BSA <0.1%
    Chelating agents (e.g. EDTA) <0.1%
    Glycerol <50%
    Sugars
    Gelatin <0.1%
    Tris <50mM
    Borate buffer
    Buffer components & conditions should not include the following:
    Antibody in ascites fluid, serum, hybridoma or tissue culture media
    Glycine / Thimerosal / Thimerosal / Merthiolate / Proclin
    Nucleophilic components (Primary amines e.g. amino acids or ethylenediamine and thiols e.g. mercaptoethanol or DTT)
    Note: Individually, these components should not affect the reaction. In combination, however, the reaction may be affected.

  • Kit Storage
  • Store kit at -20°C immediately upon receipt.
  • Conjugate Storage
  • 1. Antibody conjugates can be stored at 4°C for up to 18 months. For longer storage, conjugates can be stored at -20°C in a cryoprotectant such as 50% glycerol.
    2. Optimal storage conditions for any particular conjugate must be determined experimentally and will depend on antibody stability and optimal storage conditions.
    3. Antibody conjugates should always be stored in the dark.
