- Procedure Steps
- Allow all the reagents to come to room temperature.
Dilute the antibody to a concentration of 1 mg/mL (refer to buffer compatibility table) and add 1 μL of 147 Sm Modifier reagent for every 10 μL of antibody to be conjugated. Use 11 μL or 110 μL of the antibody mix for a 10 μg or 100 μg vial, respectively.
Open the vial and pipette 11 μL or 110 μL of the antibody mixture directly onto the lyophilized material. Gently resuspend the solution by withdrawing and dispensing the liquid until it becomes clear.
Place the cap back on the vial and let the conjugation occur at room temperature (20-25°C) for 1 hour. After the conjugation time, add 1 μL or 10 μL (10 μg or 100 μg vial respectively) of 147Sm quencher Reagent. The conjugates will be ready for use after 15 minutes and purification is not necessary.
Note: It is recommended to store the conjugates at 4°C. For longer storage, the conjugate can be stored at -20°C with a cryoprotectant such as 50% glycerol.

- Product Precautions
- Before conjugating antibodies, it is important to ensure that the purified antibody is in a suitable buffer, such as 10-50 mM amine-free buffer (e.g. MES, MOPS, HEPES, PBS) with a pH range of 6.5 to 8.5. Borate buffer can also be used.
It is recommended to avoid using buffer components that are nucleophilic, as they may react with the chemicals in the antibody Labeling Kit. Compounds containing primary amines (e.g. amino acids, ethanolamine, Tris) and thiols (e.g. mercaptoethanol, DTT) should be avoided. Thimerosal should also be avoided.
Common non-buffering salts, chelating agents, sugars, BSA (< 0.1%), glycerol (< 50%), Tris (< 50 mM), and gelatin (< 0.1%) have little to no effect on conjugation efficiency and can be used without concern.
This product is intended for research purposes only and is not intended for use in human, therapeutic, or diagnostic applications.
- Kit Storage
- Upon receipt, store the kit immediately in the dark at -20°C and check individual storage requirements for each component. If the components have not been reconstituted, the kit can be stored for up to 1 year from receipt.
- Conjugate Storage
- It is advisable to store any conjugate at 4°C. For extended storage, the conjugate can be kept at -20°C with a cryoprotectant such as 50% glycerol.
- Assay Platform
- These metals are typically detected through Atomic Absorption Spectroscopy (AAS) or Inductively Coupled Plasma Mass Spectrometry (ICP-MS). The specific method used depends on the experimental setup of the researchers.
