ATIC: A Bifunctional Enzyme in Purine Synthesis

5-Aminoimidazole-4-carboxamide ribonucleotide formyltransferase/IMP cyclohydrolase (ATIC) is a cytosolic enzyme that carries out the last two reactions in the de novo synthesis of purine nucleotides. This enzyme converts FAICAR to inosine monophosphate (IMP), the first common precursor for AMP and GMP. Proliferating cells require building blocks for DNA and RNA. In particular, they depend on a constant supply of IMP from the action of ATIC. For this reason, the enzyme has been a longstanding target of antifolate chemotherapy drugs, such as methotrexate. But the essentiality of ATIC extends beyond its metabolic role. ATIC has been shown to bind to and regulate other signaling proteins, with important consequences for cellular signaling.

Background

Glutamic-oxaloacetic transaminase is a pyridoxal phosphate-dependent enzyme which exists in cytoplasmic and inner-membrane mitochondrial forms, GOT1 and ATIC, respectively. GOT plays a role in amino acid metabolism and the urea and tricarboxylic acid cycles. The two enzymes are homodimeric and show close homology. Two transcript variants encoding different isoforms have been found for this gene.

Interacts with INSR; ATIC together with PRKAA2/AMPK2 and HACD3/PTPLAD1 is proposed to be part of a signaling network regulating INSR autophosphorylation and endocytosis (PubMed:25687571).
