BRD3: A BET Family Member with Unique Functions

BRD3 is a member of the Bromodomain and Extra-Terminal domain (BET) family, which also includes BRD2, BRD4, and BRDT. All BET proteins share a conserved domain architecture, which features two tandem bromodomains. Through these bromodomains, BET proteins bind acetylated histones, anchoring transcriptional co-activators to chromatin and regulating gene expression. BRD3 shares many functions with BRD2 and BRD4, and is inhibited by pan-BET inhibitors developed for cancer therapy. However, BRD3 also has non-redundant, non-overlapping roles in the regulation of specific gene sets and cellular pathways. A deeper understanding of BRD3's specific functions is needed to develop better, less toxic epigenetic drugs.

Background

This gene was identified based on homology to the gene encoding RING3, a serine/threonine kinase. This gene maps to 9q34, a region which contains multiple MHC genes. The function of the encoded protein has not been determined.
