C1QA: The Initiator of the Classical Complement Pathway

C1q is a large, pentameric molecule built from 3 separate chains (C1QA, C1QB, C1QC) that form the “head” of the C1 complement complex. It functions as a molecular sentinel. C1q directly binds to the Fc portions of antibodies (IgM and IgG) that are “decorating” a target (think bacterium or, in the case of autoimmune disease, a self-cell). When this occurs, a conformational change in C1q “activates” the two proteases bound to the C1 complex (C1r and C1s) and the classical complement cascade is set on fire. C1q has a unique and antibody-independent function in the brain where it is used to “tag” weak or unwanted synapses for removal by microglia, a process which is dysregulated in neurodegeneration.

Background

This gene encodes a major constituent of the human complement subcomponent C1q. C1q associates with C1r and C1s in order to yield the first component of the serum complement system. Deficiency of C1q has been associated with lupus erythematosus and glomerulonephritis. C1q is composed of 18 polypeptide chains: six A-chains, six B-chains, and six C-chains. Each chain contains a collagen-like region located near the N terminus and a C-terminal globular region. The A-, B-, and C-chains are arranged in the order A-C-B on chromosome 1. This gene encodes the A-chain polypeptide of human complement subcomponent C1q.
