CAPN2: A Calpain Isoform in Cell Migration and Fibrosis

CAPN2, also known as Calpain-2, is a highly expressed, calcium-dependent cysteine protease that assembles with the CAPNS1 regulatory subunit, similar to CAPN1, to form an active heterodimer. It shares many of the same substrates as CAPN1, but it is more selectively activated by higher calcium concentrations. Calpain-2 has unique and important roles in the regulation of focal adhesion turnover, a crucial step in the cell migration process. Calpain-2 over-activity or overexpression is closely linked to cancer cell invasion and metastasis, and tissue remodeling in cardiac fibrosis and other fibrotic disorders.
