CEACAM3: A Neutrophil Receptor for Bacterial Pathogens

Carcinoembryonic antigen-related cell adhesion molecule 3 (CEACAM3) is a type I transmembrane glycoprotein that is uniquely and highly expressed on the surface of neutrophils (or granulocytes). In contrast to other CEACAM family members, CEACAM3 does not mediate cell-cell adhesion. Rather, it is an opsonin-independent phagocytic receptor and contains an immunoreceptor tyrosine-based activation motif (ITAM) in the cytoplasmic tail. CEACAM3 directly recognizes and binds to specific CEACAM-binding adhesins expressed on the surface of pathogenic bacteria including Neisseria gonorrhoeae and Haemophilus influenzae. Binding of these bacteria by CEACAM3 elicits a strong signaling response that leads to the engulfment and destruction of the microbe.

Background

This gene encodes a member of the family of carcinoembryonic antigen-related cell adhesion molecules (CEACAMs), which are used by several bacterial pathogens to bind and invade host cells. The encoded transmembrane protein directs phagocytosis of several bacterial species that is dependent on the small GTPase Rac. It is thought to serve an important role in controlling human-specific pathogens by the innate immune system. Alternatively spliced transcript variants have been described.
