CLU: A Multifunctional Extracellular Chaperone

Clusterin, also known as Apolipoprotein J, is a highly conserved glycoprotein and one of the most abundant secreted proteins in the body. It acts as an extracellular chaperone that can associate with misfolded or stressed proteins to block their aggregation and promote clearance. Clusterin’s disease association is complex and context dependent. As a major genetic risk factor for Alzheimer’s disease, it localizes to amyloid plaques, where it likely attempts to counteract protein aggregation. Clusterin is a key mediator of the cancer cell stress response and becomes upregulated in response to therapy (chemo, radiation), where it acts as a pro-survival factor in the tumor microenvironment to prevent apoptosis.

Background

The protein encoded by this gene is a secreted chaperone that can under some stress conditions also be found in the cell cytosol. It has been suggested to be involved in several basic biological events such as cell death, tumor progression, and neurodegenerative disorders. Alternate splicing results in both coding and non-coding variants.
