CUBN: The Intestinal and Renal Absorptive Receptor

Cubilin, also known as CUBN, is a large peripheral membrane protein that serves as a major absorptive receptor in both the small intestine and kidney proximal tubules. CUBN has no transmembrane domain and is internalized through its co-receptor, Megalin (LRP2). Megalin and Cubilin bind and internalize many important ligands, most notably the Vitamin B12-Intrinsic Factor complex (required for B12 absorption). The CUBN/Megalin complex also reabsorbs filtered proteins from the urine in the kidney and prevents them from being excreted. Mutations in the CUBN gene lead to Imerslund-Gräsbeck syndrome, an inherited disease that causes vitamin B12 deficiency and proteinuria.

Background

Cubilin (CUBN) acts as a receptor for intrinsic factor-vitamin B12 complexes. The role of receptor is supported by the presence of 27 CUB domains. Cubulin is located within the epithelium of intestine and kidney. Mutations in CUBN may play a role in autosomal recessive megaloblastic anemia.
