A common technique in protein purification involves genetically engineering a 6x histidine tag onto a target protein. This string of six histidine residues exhibits a strong affinity for metal ions, particularly nickel. Immobilized-metal affinity chromatography (IMAC) exploits this interaction, allowing for the selective isolation of the tagged protein.
