LAG3: An Immune Checkpoint Receptor in Cancer Immunotherapy

LAG3 (Lymphocyte-activation gene 3) is an immune checkpoint receptor found on activated T cells, B cells, and NK cells. It helps control immune activity by suppressing T cell responses and supporting immune balance. By interacting with MHC class II molecules, LAG3 downregulates T cell proliferation and function. Due to its immunosuppressive properties, LAG3 has become a key target in cancer immunotherapy and autoimmune disease research, with therapeutic strategies focusing on blocking its inhibitory effects to enhance anti-tumor immunity and restore immune balance.

Lymphocyte-activation protein 3 belongs to Ig superfamily and contains 4 extracellular Ig-like domains. The LAG3 gene contains 8 exons. The sequence data, exon/intron organization, and chromosomal localization all indicate a close relationship of LAG3 to CD4.
